Via taxi: it takes about 25 minutes from the airport and about 20 minutes from either main station.

The Rudolf is right in the centre of Prague. You can walk to most sites from here or take a nearby tram. You can stroll along the Vltava River or enter the nearby park for splendid panoramic views of Prague.
